Context: The observation of a cell population phenotype, once it has
been disturbed, is now a major investigation tool in biology. For
instance, blocking a gene with a RNAi may highlight this gene effect
over the cell normal functioning. The observation of a toxic molecule
may show new action modes and put on the right track to develop new
medicines. As the phenotype modifications are sometimes extremely
subtle, it is necessary to develop specific fluorescent markers and to
resort to sensitive fluorescence microscopy to observe the phenotypes;
in addition, sophisticated image and data processing methods to
analyse et quantify the phenotype are to be developed. The successful
candidate for this post-doc position will contribute to develop such
methods. This study is carried out within the framework of a set of
other research projects led by the Ecole des Mines together with
several academic and industrial partners, with applications in cancer
and toxicology research.

Aims: The ideal candidate is self-motivated, holds a PhD, is able to
turn theoretical concepts into operational methods and appreciates
multidisciplinary studies, closely linked to biology. Indeed, (s)he
will develop new methods in order to :
· Extract and segment nuclei, cells and sub cellular constituents from
the information available in the multiple fluorescent channels
associated to the various markers. As cells are objects with very
specific shapes, model based segmentation has to be envisaged.
· Extract parameters to characterize the size, shape and texture in
order to identify the normal and abnormal shapes of the various cell
components. · Study the cell distribution in space, in order to
characterize the interactions among cells.
· Study the motion of living cells, by developing algorithms for
tracking them and analysing their deformation.
· The tools will be adapted to the different possible cell culture
protocoles: cell proliferation, cell differentiation, co-cultures
mixing two different cell types.

Skills and knowledge required :
· Good knowledge in image processing, mathematical morphology, and in
cell biology. Some experience of data classification and analysis
would be an additional advantage
· Good skills in programming in C and C++.
· Good knowledge of written and spoken English.

This post-doctorate position is immediately available and the initial
appointment is for one year.
